

MarathonRT® purified from Eubacterium rectale is a first-generation, processive reverse transcriptase used to create cDNA from any RNA template.

- Optimized Specific Activity: Each unit (U) of MarathonRT is defined as the amount of enzyme that incorporates 1 nmol of dTTP in 30 minutes at 42 °C with poly(rA)•oligo(dT)18 substrate
- Kit-Specific Enzyme Content: The 20-reaction kit contains 400 U total enzyme, provided at 20 U/µL in a convenient 20 µL aliquot—sufficient for robust first-strand synthesis in 20 standard reactions (1× RT mix)
- Highly-Processive cDNA Synthesis: MarathonRT consistently produces ≥90% full-length cDNA products up to 8 kb, with only a modest drop on 9.6 kb templates
- Fast and Synchronous Reverse Transcription: Copies long, structured RNAs at a constant speed of ~25 nt/s without pausing at stable RNA motifs
- Sensitive Detection of RNA Modifications: Compatible with DMS-MaPseq and SHAPE-MaP, enabling direct probing of RNA secondary structure across full-length transcripts.
- Structure-Guided Design: Crystal structures of the E. rectale maturase RT domain reveal a unique α-loop and RT0 motif that mediate high processivity
